We investigated the correlation of the expression of p53 and Ki-67 proteins in the clinical and histopathological types of seborrheic keratosis(SK). Forty-two cases of SK, including 18 acanthotic, 11 hyperkeratotic, 6 adenoid, 5 clonal and 2 irritated types, and 5 normal elderly facial control skin samples were examined by immunohistochemical staining. The number of cells that stained positive for p53 or Ki-67 was counted, and the ratio of this number to the total epidermal area, excluding the stratum corneum, was defined as the p53 or Ki-67 rate(Number/mm2). The epidermal area, excluding stratum corneum, was estimated by the Videotest/Master morphology program¨Þ (IMTechnology, Daejeon, Korea). The p53 rate was highest in the hyperkeratotic type of SK, followed in order by the clonal, acanthotic, irritated and adenoid types. The expression of Ki-67 protein was also highest in the hyperkeratotic type. However, the increased rate was statistically significant only for p53 in the hyperkeratotic type. Hyperkeratotic SKs are mainly located in sun-exposed areas, such as the face. It could be inferred from these data that the high expression of p53 in the hyperkeratotic SKs is connected to ultraviolet damage from the sun. Thus, our results suggest the different expression levels of p53 and Ki-67, according to the clinical and histological types of SK, can be interpreted as resulting from the different pathogenic mechanisms for the genesis of the specific types of SK.
